乳酸菌素Gassericin T基因的合成及其在大腸桿菌中的融合表達

[摘要] 目的:在大腸桿菌系統中表達有抗菌活性的乳酸菌素Gassericin T。方法:根據乳酸菌素Gassericin T的基因序列,
把Gassericin T的結構基因gatA編碼的氨基酸的密碼子轉換成大腸桿菌偏愛的形式;用人工合成的寡核苷酸片段,通過重
疊PCR法擴增得到gatA片段(倒基因);將合成的 基因插入pGEX一4T一1,構建pGEX一4T一1一gat融合表達載體,轉化大
腸桿菌DH5a株,IPTG誘導表達,經超聲裂解后獲得包涵體蛋白,經溶解、變性、復性處理后獲得GST-Gassericin T融合蛋
白;用瓊脂擴散法測定其對金黃色葡萄球菌、大腸桿菌、李斯特菌、枯草桿菌等的抗菌活性。結果與結論:采用pGEx一4T一1
融合表達系統在大腸桿菌中表達了有活性的Gassericin T,融合蛋白以包涵體形式存在。復性的融合蛋白對金黃色葡萄球菌
和大腸桿菌有明顯的抑制作用,對李斯特菌的抑制作用不明顯。

[Abstract]Objective:To express the bacteriocin Gassericin T with antimicrobial activities in E.coli.Methods:DNA frag—
ment encoding the mature peptide of Gassericin T was prepared by three rounds of PCR from a total of six 59 nt oligoes
after modification of the original sequence to the optional codon usage of E.coli and then cloned into pGEX T一1.Clones
carrying pGEX一4T一1一gat was induced by IPTG.The bacteria were bmken down by ultrasonic and the inclusion bodies
obtained underwent denaturation,renaturation and puriifcation to obtain GST-gassericin T fusion protein.The antimicrobial
activity of the fusion protein was assayed by agar diffusion method.Results: Gassericin T was expressed as GST—fusion
protein in pGEX一4T一1 prokaryotic expression system.The fusion protein was present in the lysate as inclusion bodies.
Gassericin T demonstrated antimicrobial activities against gram-positive.s. re and against gram—negative E.coli. Con-
clusion:Recombinant Gassericin T with antimicrobial activities was generated in pGEX一4T一1 fusion expression system.

細菌素是細菌在代謝過程中通過核糖體合成機制產生的一
類具有抑菌活性的多肽或蛋白質組分,產生菌對其細菌素有自
身免疫性【l,21。乳酸菌素是由乳酸菌產生的細菌素,其抑菌范圍主
要是與產生菌種屬相近的其他細菌【3-5],但也有一些革蘭陽性菌
Gassericin T為疏水性肽,只有C端為親水性,二硫鍵的形成可
能與其活性有關,但對其作用機理及遺傳機制都不是很明確。為
了研究Gassericin T作用的分子機制,我{flAre合成了gatA片
段, 并在大腸桿菌中高效表達了GsT—Gassericin T融合蛋白。
融合表達產物經復性后獲得了有抗菌活性的蛋白。
產生的細菌素對革蘭陰性菌起抑制作用。乳酸菌已被公認為安
全無毒的細菌。在過去的幾年里,人們對乳酸菌細菌素進行了廣
泛的基礎和應用性研究,主要是因為它們能有效地抑制食品及
1材料和方法
1.1材料
其產品中的病原菌,是潛在的生物防腐劑和飼料添加劑,在食品
和飼料的發酵和儲藏過程中起著重要的作用。近年來,隨著乳酸
菌素的不斷發現,相關的研究也越來越多。乳酸菌素Gassericin
T是一種由加氏乳桿菌(Lactob illus gasseri)SBT2055分泌的廣
表達質粒pGEx一4T_1、大腸桿菌DH5a株、金黃色葡萄球菌
ATCC6538株、鏈球菌、沙門氏菌等標準菌株均由本實驗室保存。
譜抗菌的細菌素閻,其相對分子質量為5 400,含有6個ORF。其
中第4個ORF gatA基因(簡稱gat基因)編碼活性蛋白。gatA編
碼的蛋白由75個氨基酸殘基組成,分子內含2個Cys殘基,可
形成分子內的二硫鍵,在第17和第18位有1個GG基序的切割
限制性內切酶、 DNA聚合酶為TaKaRa公司產品;丙烯酰
胺、N,N2一亞甲雙丙烯酰胺、過硫酸胺、SDS、DTT、溴酚藍、考馬斯
